Action of Pyrimethamine: A schizonticide for slow-acting blood. Action Process The thymidylate synthesis of the malaria parasite is hindered by pyrimethamine because it inhibits the enzyme dihydrofolate reductase (DHFR). An early step in the production of thymidylate, sulfadoxine inhibits dihydropteroate synthase.When taken orally. Use in Clinical Settings For the treatment of falciparum malaria, it is used in combination with sulfadoxine tablets. Potential Side Effects Rashes, allergic alveolitis, gastrointestinal problems, mild hypochromic anemia, and other symptoms.
