Pharmacology -Flurouracil
Action of Fluorouracil Interferes with the production of dTMP and thus with DNA synthesis. Mode of Action Gives rise to a counterfeit nucleotide and Inhibits thymidylate synthetase. Given i.v. Considering i.v. Clinical Use Cancers of GIT (gastric, colorectal ), pancreas, breast; malignant skin diseases. Negative Outcomes Myelosuppression, gastrointestinal problems, and mucositis are uncommon. Given long term: desquamation of feet & hands. Folic acid, a tetrahydrofolate derivative, should be used as a "rescue" after high-dose regimens to reduce harmful effects on the GIT mucosa and bone marrow.
